I have a 1970 440-6 4 speed Road Runner with air grabber and manual brakes, my question is at the rear of the intake manifold they have two nipples with hoses attached. One is going to the vacuum canister for the air grabber and the other hose is coming from the drivers side valve cover. I was told the hose from the drivers side valve cover should be going to the center carb and that I should only have the air grabber hose connected to the intake manifold. Can anyone verify what is correct for a factory 440-6 setup on this car? Thanks.....

What you've been told is correct. The PCV valve on the driver's valve cover wraps all the way around the engine & connects to the nipple at the base of the center carburetor.

For your car, you should leave the air grabber hose hooked up to the little nipple & the big nipple should have a rubber cap on it. If you had power brakes, the big nipple would connect to the power brake booster.
